PT 141 (Bremelanotide) is a synthetic cyclic heptapeptide and a melanocortin receptor agonist derived from α-MSH. It addresses sexual dysfunction by targeting central MC3R and MC4R pathways. In men, it enhances erectile function by promoting relaxation of the corpus cavernosum smooth muscle. In women, it increases sexual desire through central nervous system–mediated regulation of libido. Unlike traditional PDE5 inhibitors, PT 141 operates independently of the nitric oxide pathway, making it effective in cases resistant to PDE5 therapies. The peptide has shown a rapid onset of action, often within minutes of administration. Preclinical and clinical studies indicate that PT 141 is generally well-tolerated.
